В качестве модельных использовали природные штаммы М1062 (Астрахань, 1970 г.) и М1509 (Москва, 2012 г.). Штаммы культивировали на агаре Луриа-Бертани при температуре 37 °C. Электрофорез проводили по методу W.K. Laemmli (1970 г.), масс-спектрометрическое профилирование – согласно методу, описанному A. Shevchenko et al.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Масс-спектрометрическое сканирование лизатов клеток исследуемых штаммов показало значительное сходство их протеомов (615 общих белков). Выявленные различия касались повышенной экспрессии у штамма М1062 белков, которые участвуют в биосинтезе ДНК /РНК , входящих в группу «пурины, пиримидины, нуклеозиды», а также регуляторных белков. У штамма М1509 увеличен биосинтез белков, ответственных за патогенез, адаптацию при действии неблагоприятных факторов внешней среды, входящих в группу «кофакторы, витамины, пигменты», участвующих в липидном, углеводном, белковом обменах, клеточных процессах, и белков-транспортеров. Высказано предположение, что широкое распространение геновариантов Эль Тор, возможно, обусловлено повышением патогенных и адаптивных свойств, а также существенной перестройкой метаболизма клеток.</p></sec></abstract><trans-abstract xml:lang="en"><p>Objective of the study was to identify the differences in the production of proteins in typical strain and genetically altered variant of V. cholerae O1, biovar El Tor.</p><sec><title>Materials and methods</title><p>Materials and methods. Natural strains M1062 (Astrakhan, 1970) and M1509 (Moscow, 2010) were used as model strains in this work. Strains were cultivated on Luria Bertani agar at 37 °C. Electrophoresis was performed in accordance with W.K. Laemmli technique (1970), mass-spectrometric profiling – the method described by A. Shevchenko et al.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. Mass-spectrometric scanning of cell lysates of the examined strains showed significant similarity of their proteomes (615 common proteins). The identified differences pertained to high expression of proteins in the strain M1062, participating in biosynthesis of DNA/RNA, included into “purines, pyrimidines, nucleosides” group, as well as regulatory proteins. In M1509 strain, biosynthesis of the proteins responsible for pathogenesis, adaptation under the influence of unfavorable environmental factors, included into “co-factors, vitamins, pigments” group, involved in lipid, carbohydrate, and protein metabolism, cellular processes, as well as proteins-transporters was increased.
