Экспериментальное исследование влияния низкой температуры и дефицита питательных веществ на стабильность комплекса генетических локусов Vibrio cholerae Еltor.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В экспериментальных микрокосмах исследовалась стабильность комплекса генетических локусов (основные гены патогенности; участки генома, содержащие вариабельные тандемные повторы и сайты узнавания редкощепящих рестриктаз, определяющие VNTR- и PFGE-профиль изолятов соответственно; гены жизнеобеспечения холерного вибриона) четырех штаммов Vibrio cholerae Еltor на разных этапах инкубации.</p></sec><sec><title>Результаты и выводы</title><p>Результаты и выводы. Установлено, что при культивировании исследуемых штаммов в условиях дефицита питательных веществ и низкой температуры произошло формирование клонов с измененным VNTR- и PFGE-профилями, частота обнаружения которых составила 4–8·10–2. Изменений в структуре генов «домашнего хозяйства» и утраты генов патогенности в составе мобильных генетических элементов не выявлено. Результаты исследования доказывают вероятность изменения анализируемых локусов генома V. cholerae в период пребывания микроорганизма в неблагоприятных условиях среды, что следует принимать во внимание при интерпретации результатов молекулярного типирования.</p></sec></abstract><trans-abstract xml:lang="en"><p>Objective is an experimental study of the low temperature and nutrient deficiency effects on the stability of Vibrio cholerae El Tor genetic loci complex.</p><sec><title>Materials and methods</title><p>Materials and methods. The stability of the genetic loci (i.e. the basic pathogenicity genes; genome fragments containing variable tandem repeats and sites of rare cutting restrictase recognition defining VNTR- and PFGE-profiles of the isolates, accordingly; V. cholerae «housekeeping» genes) of the four Vibrio cholerаe El Tor strains was investigated on the experimental microcosm at different stages of incubation.</p></sec><sec><title>Results and conclusions</title><p>Results and conclusions. The study demonstrated that clones with altered VNTR- and PFGE-profiles (the detection frequency was 4–8·10–2) were being formed when strains were cultivated under nutrient deficiency and low temperature conditions. Neither changes in the structure of «housekeeping» genes nor loss of pathogenicity genes of the mobile genetic elements were revealed. The findings affirm the probability of analyzed V. cholerae loci alteration occurrence under unfavorable environment conditions.
