Методом проточной цитофлуориметрии и при регистрации тотальной флуоресценции выявлена гетерогенность популяции по размерам частиц и содержанию в них нуклеиновых кислот. Показана корреляция между концентрацией колониеобразующих единиц в суспензии и приростом флуоресценции после добавки цетилтриметиламмонийбромида. Предложено использовать этот подход в качестве экспресс-метода оценки концентрации жизнеспособных клеток в лиофилизированных/регидратированных препаратах в процессе приготовления и хранения музейных штаммов, сухих живых вакцин и пробиотических препаратов грамотрицательных бактерий.</p></abstract><trans-abstract xml:lang="en"><p>Investigated was ethidium bromide interaction with the cells of avirulent Y. pestis and Y. pseudotuberculosis strains in lyophilized/rehydrated preparations. The population heterogeneity regarding particle size and concentration of nucleic acid was identified by flow cytofluorimetry and registration of total fluorescence intensity. Shown was the correlation between concentration of colony-forming units (CFU) and fluorescence increase after supplementing suspension with cetyltrimethylammoniumbromide.
